Abstract
Poorly soluble drugs have lower bioavailability and patient compliance than highly soluble drugs. Therefore, problems such as overdose and side effects have been reported. Dexibuprofen (DP), which is a poorly soluble drug, has been used as a representative analgesic and anti-inflammatory agent. It is reported that there is a limitation in absorption in the human body after administration. DP was selected as a model, and the objective was to improve bioavailability and patient compliance by fabricating a fibrous matrix. We expected a reduction of side effects and the enhancement of therapeutic effects by mixing vitamin C (VC) with dexibuprofen. After improving the flowability and compositional uniformity of the mixture by applying the wet granulation (WG) process, a sucrose-based fibrous matrix was fabricated using the centrifugal melt-spinning process. These were composed as Batch (BAT) 1–3 depending on the presence and type of polymer binder. In this study, we fabricated a fibrous matrix composed of sucrose, polymer, dexibuprofen, and vitamin C through the centrifugal melt-spinning process, and assessed the structural changes and matrix effects in the process.
