A novel gold-based molecular beacon for probing CNG DNA repeat sequences

Abstract

We designed and developed a novel gold-based molecular beacon that targets CAG and CTG DNA repeat sequences. In single-stranded closed-loop state, the gold-based CNG repeat probe is quenched by an attached gold nanoparticle. Upon encounter with its target CAG or CTG repeat sequence, the probe forms a linear duplex structure and exhibits increased emission signal. This is the first gold quencher-based molecular beacon targeting the CNG repeat.
